제목 | A self-destructible nanoparticles for cancer therapy with ROS generator/scavenger |
초록 | The photochemical internalization (PCI) is one of the most promising therapy using light controlled anti-cancer strategy. The mechanism of PCI is disrupting the endocytic vesicle and release of entrapped drug from endosome to the cytosol. However, due to the lack of tumor targeting capability, their medical application has been limited. To overcome this drawback, we developed self-destructible nanoparticles (SDNs) which consist of grafted polysaccharide conjugated with photosensitizer (PS) as reactive oxygen species (ROS) generator, ROS scavenger (RS), and tumor targeting moieties. The SDNs are internalized by tumor targeting moiety-tumor receptor interaction. SDNs produce ROS that simultaneously trigger drug release and endosome escape of drug into cytoplasm by breaking the disulfide bond of the RS rings. This significant chemo-photodynamic dual system, which has great potential as an efficient delivery platform. |
