Short half-life and fast clearance of protein drugs require frequent administration for therapeutic efficiency, causing physiological burdens and bacterial infections. In this study, a novel injectable protein loaded hyaluronic acid microgel (HA microgel) via thiol-ene click chemistry was developed to achieve the local sustained therapeutic effect. The crosslinking density (0.4~12.6 kPa) and gelation time (41~165 sec) could be controlled. The size of microgel by emulsification was controlled, exhibiting various protein loading efficiency (52.86 ~ 99.02%). Moreover, in vitro protein release profile from microgels could be significantly adjusted by their size and crosslinking density. The protein drug carrier showed cytocompatibility and the released protein exhibited drug efficacy. From the results, we can suggest a promising carrier as a controlled delivery carrier for a variety of therapeutic proteins. Acknowledgements: 2018M3A9B5021319, 2019R1A6A1A11051471 |
