In vitro non-viral mediated VEGF gene transfer into HUVEC for angiogenesis using low molecular weight chitosan/DNA nano complex |

Depolymerized chitosan (DP-CS, molecular weights of 8517) was employed to investigate the DNA condensation ability as a gene carrier and a therapeutic agent for angiogenesis. DP-CS was chemically prepared with sodium nitrite in 6% acetic acid at 25℃. The molecular weight of DP-CS was determined by a gel permeation chromatography (GPC). ytotoxicity on HUVEC of DP-CS was measured by MTT assay. Scanning electron microscopy (SEM) was used to examine the morphology of the chitosan/DNA complex. Transfection efficacy and VEGF protein production were measured by β-galactosidase assay and VEGF protein kit, respectively. Angiogenesis influenced by DNA/DP-CS complex was measured by an optical microscopy as time went by. The cytotoxicity of DP-CS on HUVEC was little than that of the control by MTT. The size of DNA/DP-CS complex was within the range of 150 to 400nm. Transfection efficacy using VEGF DNA/DP-CS nano complex was 6 times than that using naked DNA. The maximum amount of VEGF protein produced by introducing DNA/DP-CS complex into HUVEC was about 580ng/ml. DP-CS is non-toxic and has good transfection efficacy. |
