제목 | Thermosensitive hydrogel for sustained release of proteins |
초록 | Commercial poloxamer 407 was reacted with diacyl chlorides to obtain multi-block poloxamer 407 derivatives, which was further purified by precipitation in the diethyl ether. The molecular weight of the synthesized multi-block poloxamer derivatives ranged from 90,000~100,000 daltons. The synthesized multi-block poloxamer 407 derivatives were dissolved in cold water to obtain temperature responsive hydrogel. The gelation temperature of multi-block poloxamer 407 hydrogel was determined with viscometer at various concentrations. In addition, the synthesized multi-block poloxamer 407 was added into the aqueous protein solutions e.g. interferon , and maintained at 4℃ to obtain clear solution containing proteins. The 15wt% aqueous solution of multi-block poloxamer 407 derivatives showed sol-gel transition at 21℃ and the gel residence duration was over one week in pH 7.4 PBS, at 37℃. The 15wt% aqueous solution of multi-block poloxamer 407 derivatives containing proteins was injected into SD rats subcutaneously to assay the plasma level of proteins with ELISA. In the rat PK study, protein solutions maintained the plasma level of proteins for only 1day, but proteins from the hydrogel were detected in plasma for 5days. |
