제목 Pegglated chitosan microsphere for nasal vaccine delivery system
초록 Chitosan is a valuable excipient for nasal drug delivery systems due to its biocompatibility, biodegradability and low cost. Also, it was reported that chitosan could induce cytotoxic T lymphocytes and macrophage activation. Polyethylene glycol (PEG) is a water-soluble polymer that exhibits properties such as protein resistance, low toxicity, and immunogenicity. In this study, pegglated chitosan microparticles (PCMs) for vaccine formulation were prepared based on ionic gelation process of tripolyphosphate, and then PCMs loaded with Bordetella Bronchiseptica Dermonecrotoxin (BBDNT), a virulence factor of a causative agent of atrophic rhinitis, were prepared for nasal vaccination. The particle sizes of PCM-BBDNT were smaller than 5 μm and had narrow size distribution with uniformity. Mechanical stability of chitosan microsphere (CM) was enhanced by pegglation of chitosan. BBDNT from PCMs was released faster than CMs. Also, much and longer TNFα from PCM-BBDNT were secreted than BBDNT itself. Furthermore, PCM-BBDNT stimulated more macrophage and showed higher response than BBDNT itself.
