Porous bone substitutes from avian eggshell biphasic calcium phosphate and eggshell membrane collagen I |

Bone is an inorganic/organic hybrid consisting of apatitic calcium phosphate and organic (mostly collagen) constituents. Eggs on the other hand is has an eggshell which is a calcium rich source and an eggshell membrane (ESM) which is mostly made up of collagen. In this study, we developed a porous scaffold from eggshells and ESM. Biphasic calcium phosphate (BCP), with HAp/TCP ratio of 7:3, was synthesized using hen’s eggshell as calcium source and phosphoric acid by precipitation method. Collagen-I (Col-I) was extracted from the ESM by acid-pepsin digestion and isolated by differential salt precipitation. XRD analysis confirmed the synthesis of BCP and SDS-PAGE followed by FPLC confirmed the isolation and purity of col-I. Porous BCP scaffolds were fabricated using sponge replica method then coated with col-I. The fabricated scaffolds have an interconnected macroporous structure with high porosity and compressive strength. In vitro tests using MC3T3-E1 cells showed that the egg-derived scaffolds are osteoconductive and osteogenic. In vivo implantation in rabbit femoral head defects also showed the bone regenerative effects of the egg-derived bone substitutes. |
