A Phosphorylated EdU for an advanced DNA labeling agent |

A novel thymidine analog, 5-ethynyl-2’-deoxyuridine (EdU), was developed as a DNA labeling agent. The alkyne group of EdU can be detected by fluorescent azide via “click chemistry”. The fluorescent azide can be easily diffused through tissues and can approach the incorporated EdU without DNA denaturation. However, the definite cytotoxicity of EdU has been reported that cells will eventually die after continuous incubation with EdU. We attempted to modify the EdU for reduced cytotoxicity and efficient incorporation into DNA. EdU which contains alkyne group for the “click chemistry” with fluorescent azide was synthesized from IdU. Hydroxyl group of phosphate was masked by bis-pivaloyloxymethyl (POM) protecting group to enhance cell membrane permeability. And bis-pivaloyloxymethyl phosphate was added to EdU. Its biological activities were compared to commercialized EdU in vitro. |
