제목 Large scale synthesis of self-assembled DNA structures for siRNA delivery applications
초록 DNA nanotechnology has been employed in various drug delivery applications especially for the intracellular delivery of siRNAs. Self-assembled DNA nanostrcutures have distictintive advantages over conventional gene delivery carriers such as facile control of particle size and shape, good biocompatibility and biodegradability. Inspired by isothermal enzymatic process of rolling circle amplification of DNA strands, our group has developed a system to prepare more than 200 fold increase of DNA nanostructures using a single-stranded circular DNA template. These systems are able to deliver various siRNA conjugates in selective cancer cells and show markedly higher gene silencing effect with a dose dependent manner.
