Despite many researches of photodynamic therapy (PDT) for the treatment of melanoma, there are strong insufficient biomedical requirements for effective generation of singlet oxygen after targeted delivery. Here, we investigated PDT of melanoma skin cancer using carbon dot - chlorin e6 - hyaluronate (Cdot-Ce6-HA) conjugates by transdermal delivery. Cdot-Ce6-HA conjugate was synthesized by the coupling reaction of diaminohexane (DAH) modified HA with the carboxylic groups of Ce6. In vitro PDT effect of Cdot-Ce6-HA conjugates was confirmed in B16F10 melanoma cells by MTT assay and live/dead assay. Furthermore, in vivo PDT by laser irradiation with Cdot-Ce6-HA conjugates resulted in complete deterioration of melanoma skin cancers. The antitumor effect was clearly confirmed by histological analysis with H&E staining and TUNEL assay for tumor apoptosis. Taken together, we could confirm the feasibility of Cdot-Ce6-HA conjugate for transdermal PDT of melanoma skin cancer. |
