제목 | Enhancement of Lipid extraction from Recombinant E. coli lysates associated with High-Pressure Homogenization Process, Flocculation and DMC/Methanol solvent system |
초록 | Efficient recovery of fatty acid from the recombinant strain, namely E. coli MG1655 disrupted cells is important in the biosynthesis of biofuels from renewable sources. In general, for the recovery of fatty acid from the cell, the cell should be disrupted by high pressure homogenizer. However, the disrupted lysate form an extremely stable nanosuspension (<200nm). Flocculation of E. coli high-pressure homogenized (02 passes, 5000 psi) cell suspensions with different flocculants were applied to improve the process of centrifugation and extraction due to very small size of the cell debris (i.e., nanoparticles) [1-2]. In this study, Cetyltrimethylammonium bromide (CTAB) was used as the flocculants. The size of the cell debris aggregates and solid recovery % of the disrupted cell were increased with increasing CTAB concentrations. The maximum solid recovery is more than 90% by centrifugation when CTAB was used at a concentration of 640 ppm. In addition, Dimethyl carbonate(DMC)/Methanol systems showed relatively good extraction power and less toxic. The optimum conditions for the recovery process were obtained and the effects of these operating parameters on the recovery of fatty acid were discussed. References [1] P.A Belter, E.L Cussler, W.S.
