Multi-drug resistant Mycobacterium tuberculosis (MDR-TB) is causing serious problems for public health. So rapid detection of MDR-TB is really important. In addition, because its mechanism is caused by gene mutation, efficient diagnosis for MDR-TB requires gene-based molecular diagnosis method. Conventional gene-based molecular diagnosis technologies are likely to cause false-negative or false-positive problems due to non-specific hybridization among used templates, probes, and primers. Also, they can detect only limited number of genes. In this study, we report gap filling ligation and ligase detection reaction coupled with high-resolution CE-SSCP (MGLAD-CE-SSCP) to amplify and detect MDR-TB. By using this, false-positive and negative results from non-specific hybridization can be minimized. Moreover, this method can rapidly and simultaneously detect MDR-TB bacteria. |
