A new pH-responsive 3D scaffold has been developed for stem cell culture using a novel technology. The paste state of poly(β-amino ester)-poly(ethylene glycol)-poly(β-amino ester) triblock copolymer in dichloromethane was shaped in the mold with fine salt particles layers at the top and bottom, followed by leaching process to forming the 3D scaffold. The prepared scaffold was characterized by scanning electron microscopic (SEM), stability evaluation in cell medium (pH 7.4) and solubility at pH 6.0. The results have indicated that the scaffold has homogeneously porous structure with pore size of 150 µm, excellent stability at culture condition that highly proper for diffusion, attachment and proliferation of cells. However, the scaffold could dissolved very well at pH 6.0 to harvesting the cells without harmful effect on the cells. Hence, the developed 3D scaffold has demonstrated as great candidate for stem cell culture. |
