Fabrication of tissue constructs from designed three-dimensional structures of cells using the layered method of cultured cell sheets could be an attractive approach for tissue engineering. Especially, rapid detachment of cultured cell sheets is very important to permits facile manipulation of the sheet and prevents functional damage. To accelerate the cell detachment from culture surface, poly (N-isopropylacrylamide) (PNIPAAm) nano-macrofiber sheet was prepared by electrospinning process, which produces continuous polymer fibers with diameters in diameters in the sub-micron range through the action of an external electric field imposed on a polymer solution. PNIPAAm sheets were produced by varing a variety of factors to investigate the influence of composition of solvent mixture, polymer concentration, applied voltage and needle gauges on fiber morphology. The morphology and surface area of the PNIPAAm substrates were investigated using SEM and BET sorption. The swelling/deswelling was measured by dynamic vapor sorption. In addition, cell sheet detachment from PNIPAAm substrates by hydrophilic/hydrophobic structure changes in response to temperature alteration were determined. |
