제목 Neurogenesis of Stem Cell Transfected using PEI
초록 Viral vector to delivery gene may induce undesirable safety problem, while non-viral vector such as cationic lipids and polymers are receiving the growing attention due to safety, easy handling and preparation or inexpensive cost. PEI with cationic charge can complexe with anionic phosphate of DNA through electrostatic interaction, resulting in a reduced size of the DNA and an enhanced stability toward DNase. Efficient delivery of foreign DNA into cells is an extremely critical step in gene therapy. Meanwhile, the tracing of neurogenesis of stem cells is important to identify the properties of stem cell.
Therefore, in this work, we examined the neurogenesis of stem cell after transfection using PEI. Transfection efficiency of rBMSC was about 10% in the measurement of flow cytometry and increased as N/P charge ratio increased. However, cell viability decreased with increasing N/P charge ratio. Successively, transfected rBMSC was differentiated to neural cell in the presence of neurogenesis media. We confirmed neural cell by NSE staining.
In conclusion, we confirmed that PEI proved to be an efficient gene delivery carriers for stem cell and in addition, could served as tracer for neurogenesis after transfection.
