제목 Copolymer based microcomposites and their application for ssDNA generation
초록 Aptamers are ssDNA or RNA that binds to wide variety of target molecules with high affinity and specificity producedby systematic evolution of ligands by exponential enrichment (SELEX). Compared to RNA aptamer, DNA aptamer is much more stable, favourable to be used in many applications. The most critical step in DNA SELEX experiment is the conversion of dsDNA to ssDNA. The purpose of this study was to develop an economic and efficient approach of generating ssDNA by using polyacrylamide microcompositions (microgels). DNA probes bearing 5′-terminal acrylamide modifications efficiently copolymerize with acrylamide monomers to form thermally stable DNA-containing microcomposits. Co-polymerization attachment is specific for the terminal acrylamide group. Stable probe-containing layers are easily fabricated on the surface of gels bearing exposed acrylic groups. Attachment can be accomplished using standard polyacrylamide gel recipes and polymerization techniques. Our technology can be applicable to ssDNA generation following the PCR amplification step in SELEX and ssDNA modification.
