Increasing the signal intensity of cancer region is essential for accurate diagnosis and surgery. However, there are more concern about stability of contrast agent. In this study, we designed macrocyclic chelator (DOTA)-based T1 contrast agent (CA) for stability. DOTA was conjugated with disaccharide to improve the water proton density by hydrogen bonding, 1H NMR & 13C NMR spectrum demonstrated the successful synthesis of CA. in addition, xylenol orange method and ICP-MS results showed that Gd3+ tightly bind to CA. In vitro paramagnetic properties of CA showed relatively high intensity compared to conventional DOTA-based CA. Also, we obtained an enhanced contrast intensity at tumor region. These results demonstrated that our CA can be used as a cancer diagnostic CA for MR imaging. |
