Osteogenic Differentiation of Human Mesenchymal Stem Cells on the Electrospun Poly-L-lactide Composite Nanofibers Incorporated with Calcium phosphate prepared by Electron Beam Irradiation |

As an alternative strategy to induce bone regeneration, various calcium phosphates have been prepared by acid-base reaction, precipitation, and microwave methods, which have successfully regulated osteogenic differentiation of the osteoblasts and human mesenchymal stem cells (hMSC) after incorporating with proper scaffolds. In this study, we synthesized calcium phosphate (CaP) with electron beam and then incorporated it with electrospun PLLA nanofibers. We obtained biphasic calcium phosphate (BCP) and hydroxyapatite (HAp) by controlling the pH under electron beam irradiation, and confirmed its properties through SEM, XRD, and FT-IR. The biocompatibility of the nanofibers was evaluated using hMSC, and the adherent cell morphology and cell viability of each sample were examined using fluorescence straining and a cell count kit-8 at different time intervals. Moreover, for the potential osteogenic market, alkaline phosphatase (ALP) activity was measured after 7 days of differentiation. |
