PCNA self-assembly and PCNA-FEN1 interaction investigated by analytical ultracentrifugation |

We characterized the self-assembly of human proliferating cell nuclear antigen (PCNA) and its interaction with human flap endonuclease 1 (FEN1) using analytical ultracentrifugation. PCNA undergoes a highly cooperative monomer to trimer assembly without dimer formation. Thermodynamic parameters for the association are dominated by a positive entropy change (+14.1 cal/mol.K). FEN1 is a monomer in solution and when mixed with PCNA shows binding to the trimeric form of PCNA only with the stoichiometry of this interaction is one PCNA trimer to one FEN1 monomer. |
