Biocompatibility evaluation and anti-adhesive effects of biodegradable PVA/Gelatin membranes |
초록 |
A continuous problem for patients in abdominal surgery is the peritoneal adhesion that follows the operations as complication. As a prevention method, the blended Polyvinylalcohol (PVA)/Gelatin (Gel) was fabricated by casting method can serve as barrier to prevent the peritoneal adhesion. Characterization and material properties of various ratios of PVA and gelatin (10/90, 30/70 and 50/50) were evaluated based on Scanning Electron Microscope (SEM) observation, contact angle measurement, FT-IR spectrum and biodegradation rate. Cytocompatibility study of PVA/Gel membranes were examined by in vitro experiments for instance, observation of L-929 mouse fibroblast cells proliferation and live/dead cell staining were done using confocal microscopy and the MTT assay. For in vivo evaluation of tissue anti-adhesion, PVA/Gel membranes and commercial products (Surgiwrap®) were implanted to the artificial defect that created between cecum and peritoneal wall in abdominal rats for 1 week and 2 weeks, and compared their tissue adhesion extents with non-implanted PVA/Gel membrane. |
