Tissue-Engineered Vascular by in vivo Culturing of Smooth Muscle Cells in Poly(L-lactide-co-ε-caprolactone) Scaffolds |

Vascular smooth muscle cells (VSMCs) were isolated from rabbit aortas, seeded in biodegradablr porous polymer scaffolds, and implanted subcutaneously in the dorsum of sthymic nude mice to tissue engineer vascular smooth muscle tissue in vivo. Poly(L-lactide-co-ε-caprolactone)(PLCL) copolymer showed a rubber-like elastic characteristic, in addition to an amorphous property and fast biodegradability. SMCs were seeded in 1×107 cells/ml densities.Before implantation, the scaffold-cell constructs were cultured in vitro for 5hour. The implants were harvested after in vivo incubation of 2, 5, 8, and 16weeks. The implants were examined SEM, histochemically by haematoxylin and eosin staining, by Masson's trichrome staining, and immunohistolochemically by SM -actin, myosin heavy chain, and CM-DiI labeling. The percent of smooth muscle extracellular matrices increased, while neovascularization decreased with an increased implantation period. The morphology of smooth muscle tissue, the expression of smooth muscle matrices in the PLCL scaffold, was similar to those in native vascular. PLCL scaffold facilitated cell seeding and promoted the in vivo formation of vascular smooth muscle tissue.
