제목 | Preparation and Characterization of Various Forms of type Ⅰ Collagen from Porcinal Small Intestinal Submucosa |
초록 | The selected submucosal layers of porcine small intesine, as isolated, is primarily and acellular extracellular matrix (ECM) material. Small intestinal submucosa (SIS) as a new material mainly composed of collagen supported early and abundant new blood vessel growth, and serve as a template for the constructive remodeling of several body tissue including skin, body wall, dura mater, urinary bladder, and blood vessels. Collagen is regarded as one of the most useful biomaterials. The excellent biocompatibility and safety due to its biological characteristics, such biodegradability and weak antigenecity, made collagen the primary resource in medical applications. We purified type Ⅰ acid-soluble collagen from porcinal SIS tissue. The collagen produced by digestion with pepsin in 0.05 M acetic acid solution and purification by salt precipitations. The purified collagen assumed the form of gel that had high viscosity. We prepared collagen films by placing collagen gel from SIS on glass tray at room temperature. Collagen sponges were prepared according to a directional freeze-drying procedure to achieve sponges with a uniform porous microstructure. In this study, we developed type Ⅰ collagen from SIS into the various forms such as gel, film, sheet and sponge for the possibility of the application of the medical devices.
