제목 | Evaluation of Gentamicin Sulfate Release from Crosslinked Small Intestine Submucosa Sponge |
초록 | Porcine small intestine submucosa (SIS) as a new material mainly composed of collagen and glycosaminoglycan has been widely used as a organ reconstruction without immunorejection responses. Chemically crosslinked SIS sponge was prepared and characterized as bio-interactive wound dressings and tissue engineered scaffolds. SIS powders were dissolved in 3% acetic acid aqueous solution for 48 hrs followed by pouring into mold and then fabricated by freeze-drying method. SIS sponge was prepared by cross-linked with 1-ethyl-(3-3-dimethylaminopropyl) carbodiimide hydrochloride (EDC) solution (deionized water : ethanol = 5 : 95) with 1, 5, 10, 50 and 100 mM concentrations for 24 hrs and lyophilized. Gentamicin sulfate was added on the fabricated SIS sponge with micropippet. Each SIS sponge was placed in a separate vial containing 20 ml of pH 7.4 PBS and maintained at 37℃ and 100 rpm, respectively. The buffer was taken out from the vials at predetermined intervals. The quantity of the released gentamicin sulfate measured by high performance liquid chromatography (HPLC). From the results, we confirmed that SIS sponge was good candidate for wound healing.
