제목 Extracellular vesicle protein based fluorescent gold nanolcusters for cellular imagingapplication​
초록 Recently, as the field of moleculardiagnostics grows, bioimaging and biosensing technologies based on fluorescentmetal nanocluster are being actively studied. Especially protein-basedfluorescence gold nanoclusters (AuNCs) are attracting attention due to theirhigh fluorescence signal and biocompatibility. We successfully synthesizedAuNCs based on extracellular vesicle (EVs) proteins. Specifically, we developedtwo different EVs based AuNCs (EV-AuNCs) with independent blue and redfluorescence emission using neutral (pH 7) and alkaline (pH 11.5) pH. Wevalidated the successful synthesis of EVAuNCs by FESEM, EDX, NTA and XPS. BothEVAuNCs were incubated with MCF, HeLa and HT29 cells, red emitting EVAuNCs werenot toxic, whereas the blue emitting EVAuNCs were toxic at high concentrations(≥5mg/ml). Additionally, we succeeded instaining cancer cell nucleus using red emitting EVAuNCs. EVAuNCs is the firststudy to imaging cells by synthesizing fluorescence nanomaterials in EVs.
