제목 | Highly efficient gene delivery system using tumor environment responsive materials |
초록 | Gene delivery has enormous potential as antitumoral therapy. In order to achieve efficient cargo delivery, nanotechnology have introduced diverse particles. But gene delivery effect is limited due to a lot of biological barriers. To overcome barriers, photochemical internalization (PCI) has recently emerged. The exact mechanism of PCI is based on destruction of extracellular or intracellular membranes by reactive oxygen species (ROS) upon laser irradiation. However, ROS mediated degradation of cargo genes have shown a limitation because genes could be altered due to photo-oxidation reaction. In this work, we established a stable photo/pH-responsive system using pH-responsive polymeric photosensitizer as a shielding material. This system is composed of cationic polymer and photosensitizer-conjugated pH-sensitive material to protect the genes from ROS during PCI-mediated gene transfection. In summary, our delivery system provided a superior tumor penetration and transfection efficacy. |
