제목 | Evaluation of Tissue Engineered Spinal Cord using Bone Marrow-derived Stromal Cells Seeded Polymer Scaffolds |
초록 | Bone marrow-derived stromal cells (BMSCs) were harvested from the femur of adult female Fischer rats. BMSCs were suspended at 2 × 107 cells/mL in a 25% Pluronic F127 solution and then polyglycolide (PGA) nonwoven was saturated with 50 L Pluronic F127 solution. Fischer rats received an implant consisting of the BMSCs suspended in the Pluronic F127/PGA nonwoven in the gap (T8 ~ T9; 5 mm) created by the spinal cord resection. For histological evaluation, the implants were removed after 4 and 8 weeks. Thin sections were cut from paraffin embedded tissue and histological sections were stained hematoxylin and eosin (H&E) staining and immunohistochemical staining for neurofilament (NF) and S-100 protein. It was observed that the effect of Pluronic F127/PGA nonwoven with BMSCs on neuroinduction are stronger than Pluronic F127/PGA nonwoven without BMSCs and blank (negative control). In conclusion, it seems that BMSCs plays an important role for spinal cord regeneration of tissue engineering area.
