Microfluidic Production of SERS-Active Microgels for Selective Molecular Analysis |

Raman spectra have been referred to as molecular fingerprints, which can be dramatically enhanced by metal nanostructures through surface-enhanced Raman scattering (SERS). Nevertheless, it is still difficult to use SERS in the direct molecular analysis of biological fluids because adhesive macromolecules passivate the metal nanostructures. We have designed monodisperse microgels containing metal nanoparticles using water-in-oil emulsion drops as a template with a microfluidic technology. Water-swollen network of hydrogel is highly permeable to small molecules while being impermeable to the macromolecules larger than its limiting mesh size. Taking this molecular-selective permeation, we can directly detect small molecules dissolved in complex biological fluids through SERS. For example, pesticide molecules dissolved in milk and egg can be detected without a significant loss of measurement precision. In addition, Aspirin spiked in whole blood can be also detected without pretreatment. |
