Immunoassay has been generally used for detection of biomolecules. However, conventional immunoassays have disadvantages in terms of high cost and a number of time-consuming steps. Herein, we suggest an on-chip colorimetric immunoassay based on target-specific assembly of gold nanoplasmonic particles (GNPs). To demonstrate the on-chip immunoassay, specially designed microfluidic chips were fabricated with polydimethylsiloxane and integrated on the glass slide patterned with antibody (or antibody-conjugated GNPs). And then target molecules incubated with the antibody-conjugated GNPs were injected into the prepared chip. We detected tiny amount of amyloid β, a representative biomarker of Alzheimer’s disease, by the color change of the chip surface based on plasmonic coupling between GNPs without an enzymatic amplification.
