제목 On-Chip Colorimetric Immunoassay of Biomarkers Based on Target Specific Assembly of Plasmonic Nanoparticles
초록 Immunoassay has been generally used for detection of biomolecules. However, conventional immunoassays have disadvantages in terms of high cost and a number of time-consuming steps. Herein, we suggest an on-chip colorimetric immunoassay based on target-specific assembly of gold nanoplasmonic particles (GNPs). To demonstrate the on-chip immunoassay, specially designed microfluidic chips were fabricated with polydimethylsiloxane and integrated on the glass slide patterned with antibody (or antibody-conjugated GNPs). And then target molecules incubated with the antibody-conjugated GNPs were injected into the prepared chip. We detected tiny amount of amyloid β, a representative biomarker of Alzheimer’s disease, by the color change of the chip surface based on plasmonic coupling between GNPs without an enzymatic amplification. We believe that the proposed on-chip colorimetric immunoassay will be applied to multiplexed and highly-sensitive detection for early diagnosis of diseases. *This research was supported by Nano·Material Technology Development Program through the National Research Foundation of Korea(NRF) funded by the Ministry of Science, ICT and Future Planning.(2009-0082580)
