Nitric oxide (NO) has attracted a growing interest because it can express the apoptotic activity for various cancers. At a high concentration level of NO, it can inhibit tumor growth. Herein, we selected S-nitrosoglutathione (GSNO) as an NO precursor. GSNO-loaded calcium carbonate (CaCO3)-mineralized nanoparticles were prepared by sequentially adding the aqueous solutions of GSNO and CO32- ions to an aqueous mixture of the block copolymers and Ca2+ ions. The CaCO3 phase of the nanoparticle effectively enveloped GSNO molecules at physiological pH. In contrast, at an endosomal acidic pH, the GSNO release was facilitated due to the rapid dissolution of the CaCO3 phase. The released GSNO from mineralized nanoparticles can be reacted with reducing agents, such as glutathione and ascorbic acid, to generate NO at the cytoplasm. |
