Nanoparticle-based delivery of nucleic acids for targeting multidrug-resistant bacteria |

The spread of bacterial infections due to the acquirement of antibiotic resistance has emerged as a serious and urgent public health problem and has been a major cause of mortality worldwide. Methicillin-resistant Staphylococcus aureus (MRSA) expresses various surface and secreted proteins that are responsible for its virulence and the resistance to antibiotics. Nanoparticle-based gene delivery systems that inhibit the gene expression could provide an alternative treatment option against bacterial infections caused by multiresistant bacteria. Layer-by-layer assembled gold nanoparticles (AuNPs) were modified with polyethyleneimine (PEI), nucleic acids and fluorescein to efficiently deliver an antisense oligonucleotide for the inhibition of the mecA gene in MRSA and a small interfering RNA designed against staphylocoagulase into MRSA S. aureus CCARM 3798.
