Three–dimensional fibrous BCP-PCL/GEL composite scaffold for BMP-2 delivery |

Recombinant human bone morphogenetic protein-2(rhBMP-2) coated biphasic calcium phosphate(BCP)-polycaprolactone(PCL)/gelain(GEL) composite fibrous scaffolds were fabricated successfully using electrospinning method. The electrospinning process to incorporate BCP nanoparticles into PCL/GEL scaffolds yielded a porous matrix and produced fibers in the diameter range from a few micros to nanometers. Then, rhBMP-2 was coated on these scaffolds for enhanced to bone inductivity. These rhBMP-2 coated BCP-PCL/GEL scaffolds were compared with BCP-PCL/GEL membranes by mechanical properties and biological behaviors. These scaffolds were characterized by scanning electron microscopy (SEM), electron dispersion spectroscopy (EDS), X-ray diffraction (XRD) and Fourier transform infrared (FTIR) spectra. The morphology of these scaffolds showed highly porous and interconnected structure after cross-linking and coating. Compared with BCP-PCL/GEL scaffold, the rhBMP-2 coated BCP-PCL/GEL scaffold exhibited excellent biocompatibility, cell proliferation and cell adhesion behavior resulted in the MTT assay (1, 3, 7days) and immunofluorescence analysis using pre-osteoblast MC3T3-E1 cells. Based on these results, rhBMP-2 coated BCP–PCL/GEL membranes could be considered as potential bone tissue engineering applications. |
