Hydroxyapatite (HA) is often used in the field of bone tissue engineering. In this study, collagen type Ⅰwas immobilized on the surface of HA disk using N-(3-dimethylaminopropyl)-N'-ethylcarbodiimide (EDC) / N-hydroxysuccinimide (NHS) as a crosslinking agent. MC3T3-E1 osteoblasts were seeded on the HA and collagen-immobilized HA (HA-Col) and cultured in alpha minimum essential medium (α-MEM) supplemented with 10% fetal bovine serum, 100 mg/ml penicillin and 100 mU/ml streptomycin in a 37℃ humidified atmosphere of 5% CO2. Osteoblasts were cultured for 4, 48, and 68 hours to examine cell attachment, viability and proliferation, respectively. Alkaline phosphatase (ALP) assay, Van Kossa and Alizarin S red staining were used to study in vitro differentiation of osteoblast MC3T3-E1 cell. The in vitro findings showed that the HA-Col had better positive effects on osteoblastic characteristics than HA. |
