제목 | Engineering of CYP153A33 to enhance ω-hydroxylation to overoxidation activity ratio in the biotransformation of medium-chain primary alkanols |
초록 | Medium-chain α,ω-alkanediols are a promising feedstock for polymer and chemical industries. Of these, α,ω-dodecanediol is a versatile chemical used as lubricants, adhesives, crosslinking agent and monomer fo polyesters, polyurethane, and polyamide. CYP153A33 from Marinobacter aquaeolei is known for its dodecane/1-dodecanol ω-specific oxidation activity. In this study, CYP153A33 was engineered to increase ω-specific oxidation activity. Based on structural understanding, several CYP153A33 variants were evaluated and P136A showed a significant increase in α,ω-dodecanediol production(7.12 mM from 10 mM 1-dodecanol) with an increased hydroxylation to over-oxidation activity ratio. Finally, the biotransformation activity of CYP153A33 P136A against medium-and long-chain primary alkanols was also evaluated. |
