제목 Effect of Inner Surface Functionalities of Multivalent Dendritic Ligands for Tumor Diagnosis
초록 Dendrimers are unimolecular species that are made by stepwise organic synthesis and purification. Of many kinds of dendrimers reported to date, polyamidoamine (PAMAM) dendrimers have been widely used for biological applications. Diagnostic nuclear medicine imaging such as single photon emission computed tomography (SPECT) uses radioactive tracers which provide superior image contrast even under microdosing conditions. In this study, we have prepared ten multivalent ligands with different inner surface groups as new dendrimer-based SPECT tracers for effective tumor diagnosis. Specifically, multiple copies of RGD peptide, a selective binder at the αVβ3 integrin receptor overexpressed in angiogenesis, were attached to the surface of the dendrimer. We envision that optimization of the inner surface functionalities of these multivalent dendritic ligands may improve tumor diagnosis by cooperativity upon binding of an RGD unit to the receptor.
