In-Vitro and In-Vivo Evaluation of Salt-Leached Porous PCL-PLLA 3-D Polymer Scaffolds for Bone Tissue Engineering Applications |

PLLA-PCL 3-D scaffolds were prepared using salt leaching method with NaHCO3 and freeze drying method aiming to produce a porous scaffold suitable for bone regeneration The scaffolds were prepared with varying ratio of polycaprolactone and poly(L-lactide) (PCL: PLLA 90:10, 80:20 and 70:30) to determine the optimal conditions for bone tissue engineering applications. The fabricated scaffolds were characterized from a microstructural, mechanical and biological point of view. Surface morphology was visualized using SEM and successful combination of two polymers was confirmed by FT-IR. Increased in porosity and pore size of the scaffold were observed with the increase in PLLA concentration. It also resulted to stronger scaffold as confirmed by compressive strength testing. Osteoblasts-like cells were cultured in the samples and the biocompatibility was assessed by cell proliferation assays using MTT, gene expression with RT-PCR and micrographs of proliferated cells on the sample by from SEM and confocal microscopy. The samples were found to be non-cytotoxic and that biocompatibility was further promoted with the increase in PLLA content. Samples were then implanted to male rabbits and were studied in vivo. Histological staining and micro-CT histomorphometry show that new bone formations were detected in the sample site containing the implants of the porous scaffolds. |
