Stimuli-sensitive polymers, of which physical or chemical characteristics changes upon pH, temperature and light, have been extensively studied to be used in the biomedical engineering and tissue engineering field. Among them the self-assemblies from pH-sensitive amphiphilic polymer have been studied as anticancer drug carrier. Recently, pH sensitive protein carrier is reported, consisted of poly(ethylene glycol)-b-poly(N’-citraconyl-2-aminoethyl)aspartamide (PEG-b-Pasp(EDA-Cit) diblock copolymer. In the study, to obtain pH sensitivity a citraconic anhydride is used, which is acid labile upon small drop of pH due to charge conversion property. However, the recent studies indicated that the usage of PEG is considered to be an obstacle to enter the tumor cell due to its high steric effect in spite of opsonization. In our study, we synthesized and characterized biodegradable poly(amino acid)-derivative with pH-sensitivity in order to achieve an enhanced intracellular delivery efficacy. |
