Sphingolipid metabolites including ceramide and sphingosine have been known as second messenger lipid for biological process of cell such as proliferation, differentiation, growth, and senescence. This signaling process is essential for normal cell cycle but however, phytosphingosine as well as ceramide have very low water solubility and therefore precipitated in aqueous media. In this study we designed a novel amphiphilic graft copolymer with poly(2-hydroxyethyl L-aspartamide) grafted with phytosphingosine (PHEA-g-PHS). The spherical micelle of 10-20nm diameter from PHEA-g-PHS was formed in aqueous media and was loaded with phytosphingosine. In order to investigate the properties as drug carrier, we loaded fluorescent dye conjugated phytosphingosine into polymeric micelle. The physicochemical properties were characterized with 1H-NMR, FT-IR, TEM, and DLS. In addition model drug loaded micelle was investigated with fluorescence spectrometer as well as UV/Vis spectrometer. |
