제목 Direct coating method of decellularized extracellular matrix on polymeric scaffold enhanced with magnesium hydroxide for renal tissue regeneration
초록 Renal diseases have been recognized as major health issues. In recent years, extracellular matrix (ECM) has been rapidly investigated and applied to the field of tissue engineering and regenerative medicine. In this study, fibroblast-derived extracellular matrix (FDM) was directly coated on three-dimensional porous poly(lactide-co-glycolide) (PLGA) scaffold incorporated with magnesium hydroxide (MH) nanoparticles. Optimized decellularization of confluent cells and MH effect resulted in the formation of a well-adhered and more preserved matrix. The FDM layer was successfully produced on each scaffold surface with reasonable decellularization rate higher than 97%. The improved biocompatibility of PLGA/MH/FDM scaffolds were confirmed by in vitro cell and in vivo animal studies. These results suggest that surface coating of FDM layer supported by MH would be effective for renal tissue regeneration.
