Hydrogel based bone substitute scaffold with autologous hair reinforcement for improved mechanical properties and biocompatibility |

In this study we make hydrogel based bone substitute scaffold for defected or damaged bone healing. As a proof of conceptual autologous implantation approach we have investigated on the effect of using human hair as a biocompatible reinforcing phase for improving the mechanical strength of the hydrogel based scaffold and its effect on the overall biocompatibility of the scaffold system. Chitosan and gelatin was used for making the hydrogel scaffold using their polyelectrolyte complex formation. The complex hydrogel was further strengthened by crosslinking with genepin. For improvement of biocompatibility further 10 wt% BCP was added with the hydrogel system. The hydrogel was freezdried and characterized for morphological, mechanical and biocompatibility. Scanning electron microscope (SEM) shows hair fibers were evenly dispersed with the fabricated scaffold. Bonding strength between fiber and matrix was evaluated for the strengthening evaluation. Using MT3T3-E1 Cells MTT assay and Live/dead cell assay was done to evaluate the biocompatibility. Cell adhesion and proliferation was evaluated by SEM and confocal imaging. Final biocompatibility was evaluated by invivo investigation using rat model for 1 and 2 months and was evaluated by micro CT and H&E staining. |
