제목 RGD-containing elastin-like polypeptide induces the β-cell proliferation and improves islet transplantation outcomes in diabetic mice
초록 Proliferation of beta cell plays important role in the maintenance of beta cell mass and function. Extracellular matrix components increase the proliferation of pancreatic β-cells. In this study, we investigated whether RGD-modified elastin-like polypeptide (REP), as a bioactive matrix, could enhance the proliferation of β-cells and protect islets from cell death. REP effectively increased the cell adhesion and proliferation of β-cell. REP increased cyclin D1and D2 levels, and decreased p27 levels. REP controlled the gene expression of insulin, PDX-1 and BETA2. When beta cell cultured on REP, the pathway of Akt and Erk were stimulated. Co-transplantation of REP-treated islets and REP beneath the renal sub-capsule in diabetic mice restored normoglycemia. Moreover, co-transplantation sites exhibited enhanced β-cell proliferation and vascularization. Thus, the REP treatment improve the proliferation, survival, and function of β-cell, and therapeutic potential of transplanted islets.
