Proliferation of beta cell plays important role in the maintenance of beta cell mass and function. Extracellular matrix components increase the proliferation of pancreatic β-cells. In this study, we investigated whether RGD-modified elastin-like polypeptide (REP), as a bioactive matrix, could enhance the proliferation of β-cells and protect islets from cell death. REP effectively increased the cell adhesion and proliferation of β-cell. REP increased cyclin D1and D2 levels, and decreased p27 levels. REP controlled the gene expression of insulin, PDX-1 and BETA2. When beta cell cultured on REP, the pathway of Akt and Erk were stimulated. Co-transplantation of REP-treated islets and REP beneath the renal sub-capsule in diabetic mice restored normoglycemia. Moreover, co-transplantation sites exhibited enhanced β-cell proliferation and vascularization. Thus, the REP treatment improve the proliferation, survival, and function of β-cell, and therapeutic potential of transplanted islets. |
