Development of tissue-engineered, vascularized tissue using biomaterials and stem cells will benefit patients suffering from cardiovascular diseases such as angina, infarction, and arteriopathy. This study was conducted to develop a bioengineering system to induce stem cell differentiation and peripheral blood vessel formation using using multiscale scaffold consisting fibrin hydrogel containing polycaprolactone nanofiber and surrounding alginate hydrogel. In the multiscale scaffold, mesenchymal stem cells and C2C12 were co-cultured on the nanofibrous structure and endothelial cells were encapsulated in the fibrin hydrogel for triple culture system. After subcutaneous implantation, newly well-developed cardiovascular tissue observed in the triple cultured multiscale scaffolds. We established a bioengineering cardiovascular tissue using triple culture system in the multiscale scaffolds can be applied in the future for bioengineering of an artificial vascularized tissue development. |
