Currently, research has explored the use of polymer/bioceramic composite scaffold to develop biomimetic structures and their application in tissue engineering has been explored. In this study, PLGA (75:25)/hydroxyapatite composite films were fabricated by the solvent-casting method to investigate the effect of various hydroxyapatite incorporation ratios (0, 5, 10, 15 wt% to the PLGA scaffold) for cell behavior. The scaffolds were fabricated by salt-leaching method. Chondrocytes were isolated from articular cartilage of a pig's hind leg. The tensile, compression, and degradation tests were used to determine the mechanical properties. The stiffness, ultimate tensile stress, and compression modulus of specimens were calculated. For cell test, MTT (Cell proliferation kit, Roche, USA), DNA (Quant-iT™ PicoGreen), and GAG (Blyscan, biodye science, UK) were measured. |
