In this presentation, an on-line combination of flow field-flow fractionation (FlFFF), multi-angle light scattering (MALS), and differential refractive index (DRI) detection (FlFFF/MALS/DRI) is introduced for the separation and MW characterization of sodium hyaluronate (NaHA), ultrahigh molecular weight aqueous polymers. FlFFF is an elution based separation technique applicable for macromolecules and particulate materials ranging from nanosized to supramicron sized according to sizes. Experiments were carried out with different NaHA products to achieve a successful size fractionation of NaHA molecules by FlFFF, and molecular weight distribution and conformation of NaHA products were both obtained by using MALS/DRI. The influences of sample filtration prior to FFF analysis, dissolution temperature, and ionic strength of carrier solution on the obtained molecular weight distribution of NaHA were examined by comparing molecular weight distribution and RMS radius. |
