The amine based lipidoid formulations for mRNA delivery |

Delivery of messenger RNAs (mRNAs) has drawn much attention since it show great advantages over conventional plasmid DNA (pDNA) delivery. We have developed new amine based lipidoid formulations for efficient delivery of mRNA. Resulting lipidoids show facile self-assembly with negatively charged mRNA through electrostatic interaction. A library of eight different amine-lipid based lipidoids has been screened by in vitro gene transfection. Amine-epoxydodecane (102-C10) show excellent delivery of mRNA and thereby express high Green Fluorescence Protein (GFP) expression in MEF cells. In addition, we synthesized mRNA-lipidoid formulations using lipidoids screened for in vivo delivery that are analyzed by luciferase mRNA expression. These results indicate that amine-lipid based lipidoid formulations have significant potential as a promising mRNA delivery carriers. |
