Middle East respiratory syndrome (MERS) is a viral respiratory disease caused by a coronavirus (MERS‐CoV). About 3-4 out of 10 MERS patients have died. A novel application of rolling circle amplification(RCA) as a versatile biosensor for MERS RNA pathogen detection is introduced. Long single strands of RCA products can be visualized by SYBR gold staining. When a biological sample containing MERS pathogen is applied, RCA of the DNA template can be achieved and can be visualized by under a UV lamp. In contrast, if there is no MERS pathogen in sample, no fluorescence signal can be seen. This assay shows high sensitivity and potent ability to detect various disease according to design of DNA template. Furthermore, this assay doesn’t need reverse transcription and process at room temperature making it possible to detect faster. It is likely that the development of this facile and sensitive MERS pathogen detection assay may relieve excessive workloads. |
