Protein drugs have attracted increasing interest because they are superior to chemical drugs in terms of specificity, therapeutic efficacy, and safety. However, due to poor cellular permeability and fragility of most proteins, intracellular delivery of proteins is challenging. In this work, we develop the calcium carbonate-based hybrid nanoparticles for efficient intracellular protein delivery. An apoptotic protein, cytochrome C (Cyt C) loaded nanoparticles were prepared by poly(ethylene glycol)-b-polyaspartate) (PEG-b-PAsp) mediated in-situ CaCO3 mineralization in the presence of Cyt C. The pH dependent dissolution behavior of hybrid nanoparticles and the pH-dependent Cyt C release kinetics were examined. The efficacy of the intracellular delivery of Cyt C and the apoptotic functions for tumor cells were evaluated. |
