Kinetic analysis of the hepatobiliary transport of ID-6105, a novel anticancer agent, by sandwich-cultured rat hepatocytes. |

Many endogenous and exogenous compounds are excreted from the body via the hepatobiliary route. The hepatobiliary transport system includes two essential transmembrane processes, uptake into hepatocytes via the sinusoidal membrane and secretion from hepatocytes via canalicular membrane. Kinetic properties and mechanisms of hepatobiliary transport of ID-6105, a novel anthracycline derivative antibiotics, were investigated in sandwich cultured rat hepatocytes. The hepatic uptake of ID-6105 in sandwich cultured hepatocytes consists of carrier-mediated transport was inhibited by ion substitution, and also inhibited by substrates for the organic anion transporting polypeptides. These results suggest that both hepatic uptake and biliary excretion of ID-6105 were mediated via carrier-mediated transporters. Sodium-taurocholate cotransporting polypeptide and the organic anion transporting polypeptides were responsible for hepatic uptake and P-glycoprotein for biliary excretion. |
