In vitro and in vivo evaluation of a novel OCNF (Oxidized Cellulose Nano-Fiber)-Silk Fibroin hemostasis agent and their effectiveness in hemostasis and wound healing. |

In this study a composite of OCNF-Silk Fibroin-thrombin were fabricated by grafting method. Thrombin in immobilized on OCNF-Silk Fibroin scaffold by EDC/NHS. The incorporation of thrombin on the surface of fibrous scaffold helps to accelerate hemostasis and wound healing. The physical properties of fabricated scaffold were examined by scanning electron microscope(SEM) and Fourier Transform Infrared Spectroscopy (FT-IR). SEM examination showed that the scaffold is highly porous. FT-IR showed that the conjugation occurred among OCNF and Silk Fibroin. The in vitro thrombin release from scaffold suggested that cumulative release of thrombin from scaffold is possible when the immobilization is successful. The water absorption and protein adsorption capacity of the scaffold is time dependent and also related to composition of the scaffold. In vitro degradation characteristics of the scaffold is related to amount of silk fibroin in the scaffold. Furthermore, in vitro experiment by studying of biocompatibility using MTT assay, cell proliferation assay using confocal fluorescence microscopy revealed that, this novel scaffold can help to fibroblast cell growth. |
