Hybrid Decellularized Electrospun scaffold with Pre-osteoblast ECM for bone regeneration |

Significant progress have been achieved in the field of bone tissue engineering in the recentdecades. Substantial number of research have been devoted towards fabrication and validation of ceramicimplantable materials for filling bone defects and regenerating long bones. However, there is still a needto develop materials specifically to address flat bone regeneration. The goal of this study was to analyzethe potential of using electrospun membranes with in-vitro incorporated pre-osteoblast extracellularmatrix (ECM) as a scaffold for regenerating flat bones. Electrospun membranes were fabricated usingpolycaprolactone (PCL) which was then cultured with either rat bone marrow stem cell (rBMSC), pre-osteoblast cells (MC3T3-E1) or a co-culture of rBMSC and osteoblast cells for two weeks. Uponcompletion of the stipulated culture period, the seeded membrane constructs were then decellularized toyield a hybrid scaffold composed of electrospun fibers and ECM. Samples from each culture system wereanalyzed using DNA/RNA quantification, collagen and sulfated glycosaminoglycans (sGAG) contentanalysis confocal and scanning electron microscopy (SEM). In vitro biocompatibility was determined bycell proliferation and immunostaining of differentiated MC3T3-E1 seeded on the decellularized and bareelectrospun scaffolds. Results show that inclusion of in vitro generated ECM improved cellularproliferation and enhanced cell differentiation though increased expression of ALM and OPN after 14days. |
