Characterization and in Vitro Biocompatibility of Liver Extracellular Matrix |

Liver regeneration requires growth factors, cells and a scaffold for optimal effectiveness. Previousstudies have noted the potential of decellularized liver (L-ECM) for tissue regeneration. Compared to epithelialorgans liver lobule lacks basement membrane and is mainly structured with a matrix consisting of fibronectinand collagen. L-ECM is also noted as a suitable substrate for immobilization of hepatocyte growth factor(HGF) which is beneficial in regenerating the hepatocytes. In this study several methods were investigated toproduce L-ECM viable for use as a biomaterial. L-ECM was prepared by decellularization of rabbit liverthrough freeze thaw method (FTM) or the use of Triton X-100 or sodium dodecyl sulfate (SDS). Materialproperties of the L-PG were observed through Fourier transform infrared spectroscopy (FT-IR), transmittedelectron microscopy (TEM), and scanning electron microscopy (SEM). Remaining, DNA/RNA, sulfatedglycosaminoglycans (sGAG) and collagen was also quantified to assess the effect of the decellularizationprocess. In vitro cell biocompatibility was tested using rat bone marrow stem cells (rBMSC), pre-osteoblastcells (MC3T3-E1, endothelial cells (CPAE) and fibroblast cells (L929). The biocompatibility and the healingability of the L-ECM was investigated by applying in liver defect of rat models |
