Evaluation of thermosensitive Tempo-oxidized nanocellulose fiber/chitosan hydrogel as an injectable adhesion barrier post-abdominal surgery |

Tempo–oxidized nanocellulose fiber (OCNF) is a natural material with many promising properties (biocompatibility, hemocompatibility etc.). In this study, we integrated OCNF at varying concentrations with chitosan and created a thermosensitive injectable hydrogel designed as a post-operative anti-adhesion material. The hydrogels underwent sol-gel transition at body temperature as a result of the interaction between chitosan and glycerolphosphate. The samples were characterized by Fourier transform infrared spectroscopy, scanning electron microscopy, MTT cell cytotoxicity, and in vivo study in rats. Results showed that the addition of OCNF increased biocompatibility of the hydrogels. Bone marrow-derived mesenchymal stem cells preferred to migrate and adhere at lower OCNF concentrations. A more porous structure was also observed with hydrogels containing OCNF, based on the subcutaneous implanted samples. Application as adhesion barrier after creation of abdominal abrasion in rats showed that the anti-adhesive property is dependent on the concentration of OCNF used. Hematoxylin and Eosin (H&E) staining and analysis also revealed that the addition of OCNF improved biocompatibillity with minimal inflammatory response observed. |
